Winogradsky Columns A) What is the metabolic process that utilizes oxygen in the first few weeks? B) What is the metabolic process that re-introduces oxygen towards the end? C) Cellulose served an important purpose in driving the metabolism of the column. What metabolic pathway did cellulose allow organisms in the column to partake in? Forgetting to add cellulose in the column means that ____________ was not produced, which means that ____________ couldn’t be turned into ___________ by anaerobic bacteria. D) H2S goes to two different zones of the column and partakes in two different types of metabolism. Fill in the following: Zone 1 and type of metabolism: Zone 2 and type of metabolism: E) These questions pertain to the microaerophilic zone of the column. What organisms are predominant in this zone? Name the general category of organisms and give me an example of 2 species that we expect to see in this region of the column. Could these organisms survive is there was an absence of organic carbon? If so, what source of carbon would they use and what type of metabolism would they use to break down that carbon source? F) A bacterial sample has been diluted 1/100, 1/50, 1/10, and then 1/10. 100 uL of the final solution has been plated. The plate reveals 75 colonies. Draw your dilution scheme and calculate titer of the original stock solution of bacteria. Be sure to show the dilution factor for each tube as well as cumulative dilution. Show how much bacteria are being transferred to each tube and designate how much sterile saline you’re adding to each tube for your dilutions. Keep your dilutions simple and don’t exceed a total of 1mL total volume in any of your dilution tubes.
